Sus antecedentes:

  • Las combinaciones multidrogas son cruciales para combatir la resistencia a los antibióticos y comprender la evolución microbiana.
  • Si bien se conocen los efectos sobre el crecimiento bacteriano, se comprende menos el impacto en la selección entre cepas sensibles y resistentes.
  • Por lo general, las drogas favorecen a los mutantes resistentes sobre los sensibles.

Objetivo del estudio:

  • Investigar el impacto de las combinaciones multidrogas en la selección diferencial entre las poblaciones bacterianas sensibles y resistentes.
  • Explorar si las combinaciones de fármacos pueden invertir la ventaja de selección típica para la resistencia.
  • Comprender las implicaciones para la evolución de la resistencia a los antibióticos.

Principales métodos:

  • Los ensayos de competencia directa se realizaron utilizando Escherichia coli resistente a la doxiciclina y sensible a la doxiciclina.
  • Se utilizaron combinaciones de fármacos hiper-antagonistas.
  • Los experimentos se llevaron a cabo en concentraciones subletales del fármaco.

Principales resultados:

  • Se descubrió que una clase de combinaciones de fármacos hiperantagonistas invierten la selección diferencial, favoreciendo a las bacterias sensibles sobre las resistentes.

Conclusiones:

  • Las interacciones farmacológicas pueden crear una característica no apreciada previamente en el paisaje del fitness de la evolución de la resistencia.
  • Existe una compensación entre la potencia de la interacción farmacológica y la presión de selección sobre las poblaciones resistentes.
  • Los hallazgos sugieren nuevas estrategias para diseñar terapias antimicrobianas que favorezcan la resistencia.
