Sus antecedentes:

  • Se sabe que la restricción dietética (RD) extiende la esperanza de vida y retrasa el envejecimiento en todas las especies.
  • La DR tiene un impacto significativo en la función endocrina en los mamíferos, pero no se ha demostrado un vínculo hormonal directo con la longevidad.
  • Los mecanismos moleculares por los cuales la DR influye en la longevidad no se comprenden completamente.

Objetivo del estudio:

  • Investigar el papel causal de las señales hormonales en la longevidad restringida por la dieta.
  • Identificar los genes específicos y las vías celulares involucradas en la extensión de la vida útil mediada por DR.
  • Para aclarar los mecanismos de comunicación entre las neuronas y los tejidos periféricos en respuesta a DR.

Principales métodos:

  • Utilizó el organismo modelo Caenorhabditis elegans para estudios de esperanza de vida.
  • Centrado en el gen del factor de transcripción skn-1 y su papel en las neuronas ASI.
  • Investigó las vías de señalización de las neuronas a los tejidos periféricos.

Principales resultados:

  • La longevidad inducida por la restricción dietética en C. elegans requiere el gen del factor de transcripción skn-1.
  • skn-1 actúa dentro de las neuronas ASI, un par de neuronas sensoriales en la cabeza.
  • DR activa skn-1 en las neuronas ASI, que luego señalan a los tejidos periféricos para aumentar la actividad metabólica.

Conclusiones:

  • El aumento de la vida útil en metazoos restringidos por la dieta depende de la señalización celular no autónoma.
  • Las neuronas ASI median la longevidad inducida por DR a través de un mecanismo endocrino que involucra a skn-1.
  • Este estudio demuestra un papel causal de una vía de señalización neuronal específica en la longevidad de la DR.
