Lesson: Translation
Translation is the process of synthesizing proteins from the genetic information carried by messenger RNA (mRNA). Following transcription, it constitutes the final step in the expression of genes. This process is carried out by ribosomes, complexes of protein and specialized RNA molecules. Ribosomes, transfer RNA (tRNA), and other proteins produce a chain of amino acids—the polypeptide—as the end product of translation.

Sus antecedentes:

  • Existen correlaciones consistentes entre las tasas de evolución de la secuencia de codificación y los niveles de expresión génica en todos los taxones.
  • Se debaten las presiones selectivas subyacentes que impulsan estas correlaciones.
  • Estudios anteriores observaron tendencias en la evolución de la secuencia, el uso de codones y los niveles de ARNm, pero no estaba clara una causa unificada.

Objetivo del estudio:

  • Para demostrar patrones conservados de covariación entre la evolución de la secuencia, el uso de codones y los niveles de ARNm en diversas especies.
  • Identificar una presión selectiva unificada que subyace a estas tendencias observadas.
  • Proponer y validar un mecanismo molecular que impulse estos patrones evolutivos.

Principales métodos:

  • Análisis comparativo de la evolución de la secuencia, el uso de codones y los niveles de ARNm en E. coli, levadura, gusano, mosca, ratón y humano.
  • Simulación evolutiva a nivel molecular para modelar el impacto de los errores ribosómicos y el mal plegamiento de las proteínas.
  • Análisis de las tendencias en los tejidos metazoares, en particular las neuronas.

Principales resultados:

  • Se observaron patrones conservados de covariación en todos los taxones estudiados, lo que sugiere una presión selectiva unificada.
  • Estas tendencias fueron más pronunciadas en los tejidos neuronales, vinculados a la sensibilidad al mal plegamiento de las proteínas.
  • Una simulación demostró que la selección contra la toxicidad de las proteínas mal plegadas, que surgen de errores ribosómicos, puede generar la covarianza observada.

Conclusiones:

  • La selección contra la toxicidad de las proteínas mal plegadas es un conductor primario de la covarianza observada entre la evolución de la secuencia, el uso de codones y los niveles de ARNm.
  • El modelo propuesto desafía la utilidad de las relaciones de sustitución sinónimo-sinónimo (Ka/Ks) para detectar la selección funcional.
  • La mala traducción puede desempeñar un papel importante en la etiología de las enfermedades neurodegenerativas.
