Chloroplast outer membrane proteins encoded by the nucleus are synthesized in the cytosol. Soon after synthesis, they bind cytosolic factors such as 14-3-3 protein and the Hsp70 chaperones that keep these precursors in an unfolded state until their translocation.

Sus antecedentes:

  • El retículo endoplasmático (ER) es fundamental para la síntesis y el plegamiento de las proteínas.
  • La exportación eficiente de proteínas desde el ER requiere maquinaria de transporte especializada.
  • Los componentes que regulan el transporte y la organización de Golgi (TANGO) eran previamente no identificados.

Objetivo del estudio:

  • Identificar y caracterizar los nuevos componentes involucrados en la exportación de ER y la organización de Golgi.
  • Aclarar el mecanismo por el cual TANGO1 facilita la exportación de cargas voluminosas, como el colágeno VII.
  • Para investigar el papel de TANGO1 en diferentes tipos de células.

Principales métodos:

  • Cribado de todo el genoma para identificar los componentes TANGO.
  • Análisis bioquímicos para determinar la localización de proteínas e interacciones de unión.
  • Experimentos de knockdown genético para evaluar el impacto funcional en la exportación de proteínas.
  • Microscopía de inmunofluorescencia para visualizar la localización de proteínas.

Principales resultados:

  • TANGO1, una proteína de membrana integral, se localiza en los sitios de salida de ER.
  • TANGO1 posee un dominio SH3 luminal que se une al colágeno VII y un dominio citoplasmático rico en prolina (PRD) que se une a las subunidades de capa COPII (Sec23/24).
  • TANGO1 knockdown inhibe la exportación de colágeno VII; TANGO1 guía la carga en los transportadores COPII sin entrar en el propio transportador.
  • TANGO1 se expresa en la mayoría de los tipos de células, excepto las células hematopoyéticas.

Conclusiones:

  • TANGO1 juega un papel crítico en la exportación ER de carga específica, en particular el colágeno VII.
  • TANGO1 emplea un mecanismo único para facilitar la carga de carga en los transportistas COPII mediante la interacción con la carga y la maquinaria COPII.
  • Los hallazgos sugieren que TANGO1 puede exportar otras cargas de manera similar a través de varios tipos de células.
