The absorbance of UV and visible (UV–visible) radiations is measured using a UV–visible spectrophotometer. Deuterium lamps, which emit UV radiation, and tungsten lamps, which produce radiation in the visible region, are used as light sources in UV–visible spectrophotometers. A monochromator or prism is used for diffraction grating, i.e., to split the incoming radiation into different wavelengths. A system of slits is used to focus the desired wavelength on the sample cell.

Sus antecedentes:

  • Las respuestas celulares al daño del ADN implican la modulación de la expresión génica, incluido el empalme del ARN pre-mensajero (ARNm).
  • El empalme alternativo dicta las variantes finales del ARN mensajero (ARNm), que influyen en el destino celular, como la apoptosis.
  • La regulación del empalme del pre-ARNm está estrechamente relacionada con la transcripción.

Objetivo del estudio:

  • Investigar los mecanismos por los cuales las células alteran el empalme pre-ARNm después del daño del ADN.
  • Para identificar eventos moleculares específicos que regulan el empalme alternativo después de la irradiación ultravioleta (UV).

Principales métodos:

  • Análisis del estado de fosforilación de la ARN polimerasa II en el dominio C-terminal (DTC) después de la exposición a rayos UV.
  • Medición de las tasas de alargamiento de la transcripción en respuesta a la irradiación UV.

Conclusiones:

  • Un mecanismo de cotranscripción vincula el daño del ADN, la modificación de la ARN polimerasa II y el empalme alternativo.
  • La hiperfosforilación de la ARN polimerasa II CTD y el alargamiento reducido de la transcripción son reguladores clave del empalme alternativo post-UV.
  • Esta vía juega un papel en la respuesta anti- o proapoptótica de la célula al daño del ADN.
