Proofreading

Synthesis of new DNA molecules is carried out by the enzyme DNA polymerase, which adds nucleotides on the daughter strand complementary to the template DNA strand. DNA polymerase has a higher affinity to add the correct base and ensures fidelity during DNA replication. Furthermore,  it exhibits proofreading activity during replication, using an exonuclease domain that cuts off incorrect nucleotides from the nascent DNA strand.

Resumen

Los "pares de bases" de colorantes catiónicos estabilizan significativamente los duplexos de ADN a través de fuerzas electrostáticas y de apilamiento. Objetivo del estudio:

  • Para investigar los efectos de estabilización de la incorporación de tintes catiónicos "pares de bases" en las ODNs.
  • Para evaluar el impacto de las interacciones electrostáticas y de apilamiento en la estabilidad del ADN dúplex.
  • Para comparar la estabilidad térmica de los ODN modificados con las secuencias nativas y las que contienen tintes neutros.

Principales resultados:

  • La incorporación de "pares de bases" de colorante catiónico p-metilstilbazole estabilizó en gran medida el dúplex de la ODN.
  • Los ODN modificados exhibieron temperaturas de fusión significativamente más altas en comparación con los pares de bases nativos y los ODN con tintes neutros.
  • El aumento del número de tintes catiónicos "pares de bases" condujo a una mayor estabilización dúplex.

Conclusiones:

  • Los "pares de bases" de colorantes catiónicos son efectivos para estabilizar duplexos de ADN a través de interacciones electrostáticas y de apilamiento.
  • Esta modificación ofrece una estrategia prometedora para mejorar la estabilidad térmica de los oligonucleótidos.
  • Los hallazgos tienen implicaciones para el diseño de estructuras y terapias estables basadas en ácido nucleico.
