Resumen

La persistencia de la memoria a largo plazo depende de la actividad específica del receptor de dopamina D1 en el hipocampo. La manipulación de este sistema en momentos críticos después del aprendizaje puede controlar la duración de la memoria.

Sus antecedentes:

  • La persistencia de la memoria a largo plazo (LTM) es una característica clave, sin embargo, los mecanismos subyacentes siguen sin estar claros.
  • Comprender los factores que influyen en la duración de la LTM es crucial para la investigación de la memoria.

Objetivo del estudio:

  • Investigar el papel de los receptores de dopamina D1 en el hipocampo en la regulación de la persistencia de la memoria del miedo a largo plazo.
  • Identificar las ventanas de tiempo críticas y las vías moleculares involucradas en la consolidación y descomposición de LTM.

Principales métodos:

  • Las ratas fueron entrenadas en una tarea de condicionamiento del miedo.
  • El antagonista del receptor de dopamina D1 (SCH23390) o el agonista (SK38393) se administró en el hipocampo dorsal en diferentes puntos de tiempo posteriores al entrenamiento.
  • Se evaluaron los niveles de factor neurotrófico derivado del cerebro (BDNF) y la regulación del área tegmental ventral (VTA).

Principales resultados:

  • La administración de SCH23390 12 horas después del entrenamiento, pero no inmediatamente ni 9 horas después, causó una rápida disminución del miedo LTM.
  • La administración de SK38393 en el punto crítico de 12 horas convirtió una memoria de miedo transitoria en una persistente.
  • Estos efectos fueron mediados por el BDNF y regulados por el VTA.

Conclusiones:

  • La persistencia de la memoria del miedo a largo plazo depende de la activación de las conexiones dopaminérgicas VTA / hipocampo.
  • La manipulación específica de este sistema dopaminérgico a intervalos definidos después del aprendizaje puede modular la duración de LTM.
