Different notations are used to represent the three-dimensional structure of molecules on two-dimensional surfaces. One of the most commonly used representations is the dash-wedge formula. Sus antecedentes:

  • El origami de ADN permite el ensamblaje preciso a nanoescala de estructuras complejas.
  • La creación de nanoestructuras huecas tridimensionales (3D) de ADN presenta desafíos en cuanto a la estabilidad y la apertura controlada.

Objetivo del estudio:

  • Diseñar y fabricar nuevas estructuras de ADN multibrazo utilizando andamios de origami de ADN 2D.
  • Para investigar el plegamiento de estos andamios 2D en estructuras 3D huecas.
  • Para analizar el mecanismo de apertura de las estructuras de ADN 3D resultantes.

Principales métodos:

  • Diseño de estructuras de ADN de múltiples brazos utilizando plantillas de origami de ADN en 2D.
  • Plegado de andamios 2D en estructuras huecas de prisma 3D a través de hilos de conexión.
  • Microscopía de fuerza atómica de alta velocidad (HS-AFM) para la obtención de imágenes en tiempo real de la dinámica estructural.

Principales resultados:

  • Diseño exitoso y plegado de estructuras de ADN multibrazo en prismas 3D huecos.
  • Las imágenes HS-AFM visualizaron el proceso de apertura de las estructuras 3D.
  • Observación de la disociación de los brazos de conexión como mecanismo para la apertura de la estructura.

Conclusiones:

  • El estudio demuestra un método para crear nanoestructuras de ADN 3D huecas conmutables.
