Sus antecedentes:

  • La proteína del nucleocáptido (NC) del VIH-1 es multifuncional y se une a los ácidos nucleicos (AN) de manera no específica.
  • La función conocida del NC implica la fusión parcial de los NA estructurados, lo que facilita la transcripción inversa del VIH-1.
  • Esta función se atribuye a la preferencia de NC por las bases de un solo hilo.

Objetivo del estudio:

  • Para investigar un modo de interacción no descubierto previamente entre la proteína NC y los ácidos nucleicos.
  • Para sondear la flexión inducida por NC de segmentos de ADN / ADN y ADN / ARN completamente duplicados.
  • Para analizar la dinámica conformacional heterogénea asociada en el modelo de complejos NC/NA.

Principales métodos:

  • Transferencia de energía de resonancia de fluorescencia de una sola molécula (SM-FRET) in vitro.
  • Utilizando modelos de complejos NC/NA para estudiar las interacciones.
  • Analizando la dinámica conformacional de los segmentos de ácido nucleico.

Principales resultados:

  • Se identificó un nuevo modo de interacción NC/NA: la curvatura aguda inducida por NC del ADN/ADN dúplex y el ADN/ARN.
  • Se observaron dinámicas conformacionales heterogéneas asociadas con la flexión inducida por NC.
  • NC demuestra una capacidad para doblar completamente los segmentos de ácido nucleico de doble cadena.

Conclusiones:

  • La proteína NC puede inducir una flexión significativa en los ácidos nucleicos dúplex, una interacción previamente desconocida.
  • Esta curvatura NA inducida por NC puede ser biológicamente relevante para la condensación mediada por NC del ADN duplex proviral.
  • Los hallazgos amplían nuestra comprensión del papel multifuncional del NC en el ciclo de vida del VIH-1.
