Sus antecedentes:

  • Los progenitores neuronales generan diversos tipos de células a lo largo del tiempo, pero los mecanismos que controlan estas transiciones temporales siguen sin estar claros.
  • La "cascada temporal de genes" en Drosophila CNS involucra factores de transcripción que alteran la competencia del progenitor.
  • No se comprende bien cómo las amplias ventanas temporales en los linajes de células madre neurales se subdividen para generar tipos celulares únicos.

Objetivo del estudio:

  • Investigar los mecanismos que subdividen las amplias ventanas temporales en los linajes de progenitores neurales.
  • Para dilucidar cómo se generan destinos celulares específicos dentro de ventanas temporales más amplias en el SNC de Drosophila.
  • Comprender el papel del gen del ricino en la progresión temporal y la diversificación del destino celular.

Principales métodos:

  • Análisis de un linaje de progenitor neural identificable en el sistema nervioso central (SNC) de Drosophila.
  • Investigando la función del gen del ricino y sus objetivos aguas abajo.
  • Identificación y caracterización de los circuitos reguladores de alimentación avanzada que controlan la expresión génica temporal.

Principales resultados:

  • Se descubrió que una amplia ventana temporal del ricino estaba subdividida por dos bucles de avance de alimentación distintos, ambos iniciados por el ricino.
  • El primer bucle de avance de alimentación especifica un destino único de la célula.
  • El segundo bucle de avance de alimentación suprime el primer bucle, permitiendo la generación de destinos celulares alternativos.

Conclusiones:

  • La progresión temporal de los progenitores neuronales está regulada por cascadas de genes que involucran genes "temporales" y "subtemporales".
  • Los bucles de avance de alimentación opuestos, activados por el ricino, subdividen amplias ventanas temporales para generar diversidad de tipo celular.
  • Este mecanismo regulador puede conservarse en varios linajes de células madre para generar diversidad celular.
