Sus antecedentes:

  • Los complejos de poros nucleares (NPC) median el transporte entre el núcleo y el citoplasma.
  • Los NPC deben ensamblarse en la envoltura nuclear (NE) durante la mitosis y la interfase.
  • Existen distintos eventos celulares y restricciones topológicas durante la reformación y el crecimiento de la NE.

Objetivo del estudio:

  • Investigar los requisitos diferenciales de las nucleoporinas durante el ensamblaje de NPC en distintas etapas del ciclo celular.
  • Para dilucidar los mecanismos que rigen el ensamblaje de NPC en el contexto de la mitosis abierta y la topología de la envoltura nuclear interfase.

Principales métodos:

  • knockdown mediado por ARN de las nucleoporinas específicas (ELYS, POM121).
  • Análisis del reclutamiento complejo Nup107/160 a la cromatina y la envoltura nuclear.
  • Investigación del papel del dominio de detección de curvatura de la membrana de Nup133 en el ensamblaje de NPC.

Principales resultados:

  • ELYS es esencial para el ensamblaje de NPC después de la mitosis, pero no durante la interfase.
  • Se requiere POM121 para incorporar el complejo Nup107/160 en nuevos sitios de ensamblaje durante la interfase.
  • El dominio de detección de la curvatura de la membrana de Nup133 es crítico para el ensamblaje de NPC de interfase, pero no para la formación de NPC postmitóticos.

Conclusiones:

  • Los NPC de metazoos utilizan al menos dos mecanismos de ensamblaje distintos adaptados a la topología NE dependiente del ciclo celular.
  • ELYS y POM121 desempeñan funciones específicas y no superpuestas en el ensamblaje de NPC durante la mitosis y la interfase, respectivamente.
  • Los hallazgos revelan adaptaciones específicas del ciclo celular en la biogénesis de NPC crucial para la integridad de la envolvente nuclear.
