Resumen

La supervivencia de la DLBCL similar a las células B activadas depende de las mutaciones de la proteína adaptadora MYD88. Sus antecedentes:

  • El linfoma difuso de células B grandes de tipo B activado (ABC) (DLBCL) es un subtipo desafiante con opciones curativas limitadas.
  • Las vías de señalización del factor nuclear constitutivo (NF) -κB y la quinasa JAK son cruciales para la supervivencia celular de las células ABC DLBCL.
  • Los fundamentos genéticos precisos de esta señalización aberrante siguen siendo incompletamente entendidos.

Objetivo del estudio:

  • Para investigar el papel de la proteína adaptadora MYD88 en la patogénesis de ABC DLBCL.
  • Identificar y caracterizar las mutaciones oncogénicas en MYD88 dentro de los tumores ABC DLBCL.
  • Para aclarar las consecuencias funcionales de las mutaciones MYD88 en las vías de señalización y la supervivencia celular.

Principales métodos:

  • Cribado de interferencia de ARN para evaluar la esencialidad de MYD88, IRAK1 e IRAK4 para la supervivencia de ABC DLBCL.
  • La resecuenciación de ARN de alto rendimiento para identificar mutaciones en MYD88.8.
  • Ensayos funcionales para evaluar el impacto de las mutaciones MYD88 en el ensamblaje del complejo proteico, la actividad de la quinasa y la señalización aguas abajo.

Principales resultados:

  • MYD88 y las quinasas asociadas IRAK1/IRAK4 son esenciales para la supervivencia de las células ABC DLBCL.
  • Se identificaron mutaciones oncogénicas recurrentes en el dominio del receptor MYD88 Toll / IL-1 (TIR) en ABC DLBCL.
  • La sustitución L265P en MYD88 se encontró en el 29% de los tumores ABC DLBCL y actúa como una mutación de ganancia de función, promoviendo la señalización constitutiva y la supervivencia celular.
  • Las mutaciones MYD88 también se observaron en otros subtipos de linfoma, incluido el linfoma de tejido linfoide asociado a la mucosa.

Conclusiones:

  • La vía de señalización MYD88 es parte integral de la patogénesis de ABC DLBCL.
  • Las mutaciones MYD88 de ganancia de función, como L265P, impulsan la oncogénesis mediante la activación de las vías NF-κB y JAK/STAT3.
  • Dirigirse a la quinasa IRAK4 y otros componentes de la vía MYD88 presenta una estrategia terapéutica prometedora para el DLBCL mutado por MYD88.
