Objetivo del estudio:

  • Investigar el reordenamiento de los complejos terminales de fibra musgosa en los circuitos del hipocampo y del cerebelo al aprender en ratones.
  • Determinar el papel funcional de estos reordenamientos estructurales en el aprendizaje y la memoria.
  • Establecer un vínculo causal entre la plasticidad sináptica inducida por el aprendizaje y la precisión de la memoria.

Principales métodos:

  • Se utilizaron modelos de ratón (Rab3a(-/-) y Add2(-/-)) para estudiar la plasticidad sináptica y la memoria.
  • Cambios examinados en las sinapsis filopodial en las interneuronas de rápido aumento después de un ensayo y el aprendizaje incremental.
  • Evaluó la precisión de la memoria y los patrones de expresión de c-Fos en el hipocampo (región CA3) después de aprender tareas como el condicionamiento del miedo contextual y el laberinto de agua de Morris.

Principales resultados:

  • El aprendizaje indujo aumentos duraderos y reversibles en las sinapsis filopodiales en las interneuronas, lo que mejora la inhibición de la alimentación.
  • El aumento de la inhibición del feedforward restringió las neuronas que expresan c-Fos durante la recuperación de la memoria y se correlacionó con la calidad de la memoria.
  • En ratones Rab3a, los cambios sinápticos inducidos por el aprendizaje y la precisión de la memoria se vieron afectados; en ratones Add2, la inhibición del crecimiento de la alimentación fue abolida, lo que condujo a conjuntos de c-Fos agrandados y memoria imprecisa.

Conclusiones:

  • Los aumentos relacionados con el aprendizaje en sinapsis específicas mejoran causalmente la precisión del aprendizaje y la memoria en los cerebros adultos.
  • La plasticidad y la inhibición del crecimiento de la alimentación en las fibras de musgo del hipocampo son fundamentales para la precisión de los recuerdos dependientes del hipocampo.
  • La restauración de la adducina 2 (Add2) en las fibras de musgo rescató tanto el crecimiento de la inhibición de alimentación avanzada como la precisión de la memoria, confirmando su papel.
