The first human genome sequencing project cost $2.7 billion and was declared complete in 2003, after 15 years of international cooperation and collaboration between several research teams and funding agencies. Agarose gel electrophoresis is very useful in separating DNA fragments by size. Running a DNA ladder containing fragments of the known length alongside the sample helps determine the approximate length of the sample DNA fragments. However, additional steps are needed to verify the sequence identity of the sample DNA fragments.

Proofreading

Synthesis of new DNA molecules is carried out by the enzyme DNA polymerase, which adds nucleotides on the daughter strand complementary to the template DNA strand. DNA polymerase has a higher affinity to add the correct base and ensures fidelity during DNA replication. Furthermore,  it exhibits proofreading activity during replication, using an exonuclease domain that cuts off incorrect nucleotides from the nascent DNA strand.

  • La ligadura en los residuos de prolina en el extremo C sigue siendo un desafío debido a la escasa reactividad de los tioésteres de prolina.

Objetivo del estudio:

  • Desarrollar una nueva estrategia para la ligadura eficiente de proteínas en los sitios N-terminales de la prolina.
  • Para superar las limitaciones de los métodos NCL tradicionales cuando se trata de residuos de prolina.

Principales métodos:

  • El estudio combinó la ligación química nativa (NCL) con la desprotección libre de metales (MFD).
  • Este enfoque fue diseñado específicamente para abordar los problemas de reactividad asociados con los tioesters de prolina.

Principales resultados:

  • Demostró la viabilidad de la ligadura en un sitio de proteína N-terminal.
