These modifications play an important role in the function and stability of these proteins. The ubiquitin-proteasome pathway is a well-known mechanism utilized by eukaryotic cells to remove cytoplasmic proteins that are misfolded, damaged, or no longer needed. Sus antecedentes:

  • Caracterizar las interacciones agua-proteína a nivel atómico es un desafío.
  • La espectroscopia de Resonancia Magnética Nuclear (RMN) de Solución ofrece potencial para el estudio de la dinámica de hidratación de proteínas, pero sufre de artefactos.
  • Métodos anteriores permitieron la cuantificación de las interacciones de las proteínas con el agua superficial utilizando los efectos Overhauser nucleares.

Objetivo del estudio:

  • Ampliar los métodos de RMN para el análisis detallado de la dinámica de hidratación de proteínas.
  • Para investigar las interacciones dipolares entre el agua de hidratación y los hidrógenos ligados al carbono de proteínas.
  • Para mapear la dinámica de hidratación a través de la superficie de la proteína.

Principales métodos:

  • Encapsulación de proteínas en partículas de micela inversa para controlar la dinámica del agua.
  • Utilizando propano líquido de baja viscosidad para manipular los tiempos de reorientación molecular.
  • La adquisición de un marco giratorio con efectos Overhauser nucleares (ROPE) utilizando (1) H unido a señales (13) C.
  • Medición de las constantes de tiempo de relajación T(1ρ).

Principales resultados:

  • Se obtuvieron datos ROPE de alta calidad alargando los tiempos de relajación.
  • Las dinámicas de hidratación se mapearon en la mayoría de la superficie de la proteína, incluidas las interacciones con hidrógenos unidos al nitrógeno.
  • Se observó un amplio espectro de dinámicas de hidratación, con agrupaciones evidentes en la superficie molecular.
  • Las regiones con agua de hidratación de larga duración se correlacionan con sitios de unión a las proteínas.

Conclusiones:

  • El enfoque de RMN extendido proporciona información detallada sobre la dinámica de la hidratación de las proteínas.
  • El agrupamiento de hidratación observado y el agua de larga vida en los sitios de interacción sugieren una optimización evolutiva de la entropía del disolvente en la unión.
  • Este método avanza en la comprensión del papel del agua en la función de las proteínas y el reconocimiento molecular.
