Sus antecedentes:

  • Las variaciones estructurales (SV) son frecuentes en los genomas de mamíferos y están implicadas en enfermedades.
  • La abundancia, los orígenes y el impacto funcional de los SV en la variación fenotípica no se comprenden completamente.

Objetivo del estudio:

  • Identificar y caracterizar exhaustivamente las variantes estructurales en cepas de ratones endogámicos.
  • Investigar las causas y consecuencias funcionales de las variantes estructurales.

Principales métodos:

  • Análisis experimentales y automatizados combinados para identificar SVs en trece cepas de ratones endogámicos clásicos y cuatro de origen silvestre.
  • Mapeó los puntos de ruptura SV a la resolución del par de bases.
  • Se analizó el impacto de los SV en la expresión génica y la variación fenotípica.

Principales resultados:

  • Se identificaron 711.920 SV en 281.243 sitios en los genomas de ratones.
  • La mayoría de los SV son pequeños (<1 kb), siendo el 98% deleciones o inserciones.
  • Las inserciones de retrotransposón representan más de la mitad de los SVs identificados.
  • Las SV tienen menos probabilidades que otras variantes de secuencia de afectar la expresión génica o los rasgos cuantitativos.
  • Identificó 24 SVs que interrumpen los exones codificantes, con un tercio que afecta a los genes inmunológicos.

Conclusiones:

  • Las variantes estructurales son abundantes en los genomas de ratones, impulsadas principalmente por la actividad del retrotransposón.
  • A pesar de su prevalencia, las SV tienen un impacto limitado en la expresión génica y el fenotipo en comparación con otras variantes.
  • Los SVs raros y de gran efecto pueden interrumpir significativamente la función génica, particularmente impactando las vías inmunológicas.
