Complex microtubule structures are present in resting cells and in dividing cells. In resting cells, they are responsible for maintaining the cellular architecture, tracks for intracellular transport, positioning of organelles, assembly of cilia and flagella. They mediate the bipolar spindle assembly for chromosomal segregation and positioning of the cell division plate in dividing cells. Intermediate filaments are cytoskeletal proteins with higher tensile strength and flexibility than microfilaments and microtubules. Unlike the other two cytoskeletal proteins, intermediate filament formation lacks the enzymatic activity to hydrolyze nucleotides like ATP and GTP to generate energy for polymerization. Therefore, the formation of intermediate filaments is multistep self-assembly. The involvement of any accessory proteins in intermediate filament formation has not yet been reported.

Objetivo del estudio:

  • Demostrar una relación cuantitativa entre el tamaño supramolecular de las nanopartículas y la energía libre de asociación.
  • Identificar las características estructurales de los tintes azoicos que influyen en la formación y el tamaño de las nanopartículas.

Principales métodos:

  • Autoensamblaje electrostático de dendrímeros de polielectrolitos catiónicos con moléculas de colorantes orgánicos.
  • Síntesis del grupo sulfonato portador de tintes azoicos.
  • La dispersión de la luz, las mediciones del potencial ζ, la calorimetría de titulación isotérmica (ITC) y la espectroscopia UV-vis.

Principales resultados:

  • Se observaron radios hidrodinámicos de 20 nm < R (((H) < 50 nm y valores positivos de potencial ζ.
  • Se requiere una ganancia mínima de energía libre de ΔG ≈ -32 kJ mol−1 para la interconexión de dendrimeros.
  • Las interacciones colorante-colorante fueron identificadas como el principal factor que controla el tamaño de las partículas.

Conclusiones:

  • Se desarrolló un modelo cuantitativo relacionado con la energía libre y el tamaño de la partícula, lo que permite la predicción basada en mediciones termodinámicas.
  • Los bloques de construcción moleculares pueden diseñarse para controlar el tamaño del ensamblaje supramolecular.
  • Este trabajo facilita el diseño dirigido de nanopartículas supramoleculares con tamaños predecibles.
