Overview

    Sus antecedentes:

    • La infección por el virus del polioma en células de ratón produce ARN específico del virus.
    • Comprender la estructura del ARN viral es crucial para descifrar los mecanismos de expresión génica.

    Objetivo del estudio:

    • Para identificar y caracterizar las estructuras con tapa en el extremo 5' del ARN tardío específico del virus del polioma.
    • Para determinar la ubicación genómica y la presencia de estas estructuras de tapa en diferentes especies de ARNm tardías.

    Principales métodos:

    • Análisis del ARN viral marcado con 32P utilizando la digestión de RNase T1, T2, A y P1.
    • Electroforesis en gel bidimensional para fraccionar las estructuras de ARN.
    • Mapeo de las estructuras de la tapa al genoma viral del polioma.

    Principales resultados:

    • Se identificaron siete estructuras principales de la capa I (m7GpppNm1pN2p) y cuatro derivados de la capa II (m7GpppNm1pNm2pN3p) en el ARN viral.
    • Estas estructuras de tapa se asignan a una región específica (66-71 unidades de mapa) en el genoma viral.
    • Todas las estructuras de tapa identificadas se encontraron en ARNm tardíos purificados 16S, 18S y 19S.

    Conclusiones:

    • Los ARNm tardíos del virus del polioma están limitados con diversas secuencias de líderes.
    • Estos líderes con tapa están unidos al cuerpo de ARNm a través de un mecanismo de empalme.
    • Los hallazgos proporcionan información sobre el procesamiento post-transcripcional del ARNm viral.
