Labeling DNA Probes

DNA probes are fragments of DNA labeled with a reporter tag to enable their detection or purification. The resulting labeled DNA probes can then hybridize to target nucleic acid sequences through complementary base-pairing, and may be used to recover or identify these regions.

Sus antecedentes:

  • El cribado fenotípico identifica pequeñas moléculas bioactivas, pero la identificación del objetivo es difícil debido a los efectos fuera del objetivo.
  • La caracterización de las interacciones de pequeñas moléculas en los sistemas vivos sigue siendo difícil, lo que dificulta la comprensión mecánica de la actividad farmacológica.

Objetivo del estudio:

  • Desarrollar una biblioteca de moléculas pequeñas novedosa y completamente funcional para la identificación acelerada de objetivos.
  • Crear una herramienta para visualizar e identificar objetivos proteicos de compuestos bioactivos in situ.

Principales métodos:

  • Se diseñó una biblioteca de moléculas pequeñas con elementos de diversidad, un grupo fotorreactivo para el enlace cruzado y un mango de alquina para la conjugación de reporteros.
  • Se emplearon químicos quimioproteómicos cuantitativos y ensayos in vitro para identificar y validar el objetivo de un compuesto activo.

Principales resultados:

  • Un miembro de la biblioteca inhibió selectivamente la proliferación de células cancerosas en condiciones de baja glucosa.
  • La quimioproteómica cuantitativa identificó a la NADH:ubiquinona oxidorreductaza (complejo 1) como el objetivo específico.
  • El compuesto inhibió la actividad del complejo 1 in vitro, consistente con la muerte celular observada bajo limitación de nutrientes.

Conclusiones:

  • Las bibliotecas de compuestos completamente funcionales junto con la quimioproteómica permiten una rápida identificación de objetivos y caracterización mecánica.
  • Este enfoque acelera el descubrimiento de sondas bioactivas para procesos biológicos complejos.
