Resumen
La vía SUV39H1H3K9me3HP1α mantiene la estabilidad de las células T ayudante 2 (TH2) al silenciar los genes T ayudante 1 (TH1). La pérdida de esta vía promueve las respuestas TH1 y exacerba el asma alérgica.

Sus antecedentes:
- Las células T CD4+ naïves se diferencian en subconjuntos especializados de T auxiliar (TH) (TH1, TH2, TH17, Treg) cruciales para la inmunidad.
- Los mecanismos epigenéticos regulan la diferenciación de las células TH, pero su papel en la estabilidad del linaje sigue sin estar claro.
- La vía SUV39H1H3K9me3HP1α está involucrada en el silenciamiento de la transcripción a través de la formación de heterocromatina.
Objetivo del estudio:
- Investigar el papel de la vía SUV39H1H3K9me3HP1α en el mantenimiento de la estabilidad del linaje de T helper 2 (TH2).
- Para determinar si esta vía influye en el compromiso de las células TH2 con otros linajes.
Principales métodos:
- Se estudiaron las células TH2 deficientes en SUV39H1 o HP1α.
- Se evaluaron las modificaciones histónicas (H3K9me3, H3K9ac) y la unión a HP1α en los loci del gen TH1.
- Expresión génica analizada de las células TH1 y TH2 bajo condiciones inductoras de diferenciación.
- Utilizó un modelo de ratón de asma alérgica impulsada por TH2 para evaluar la relevancia in vivo.
Principales resultados:
- La deficiencia de SUV39H1 deterioró las relaciones de trimetilación / acetilación de H3K9 y redujo la unión de HP1α a los promotores silenciados del gen TH1 en las células TH2.
- Las células TH2 con deficiencia de SUV39H1 o HP1α expresan anormalmente los genes TH1 en el cultivo inductor de TH1.
- In vivo, la inhibición o pérdida de SUV39H1 desplazó las respuestas de las células T hacia TH1 y redujo la patología del asma alérgica.
Conclusiones:
- La vía SUV39H1H3K9me3HP1α es esencial para mantener la estabilidad del linaje TH2 al silenciar epigenéticamente los loci TH1.
- Esta vía representa un objetivo terapéutico potencial para enfermedades inflamatorias mediadas por células TH2 como el asma alérgica.
