Sus antecedentes:

  • La pluripotencia de las células madre embrionarias (ES) se estudia principalmente a través de la transcripción, la cromatina y el ARN no codificante.
  • El papel del empalme alternativo en la pluripotencia y diferenciación de las células ES sigue siendo en gran medida inexplorado.

Objetivo del estudio:

  • Investigar el papel del empalme alternativo en la regulación de la pluripotencia y la diferenciación de las células ES.
  • Identificar los reguladores clave del empalme alternativo en las células ES.

Principales métodos:

  • Identificó proteínas de tipo muscular ciego (MBNL) (MBNL1 y MBNL2) como reguladores del empalme alternativo.
  • Examinó los efectos de la eliminación de la proteína MBNL en las células diferenciadas y la sobreexpresión en las células ES en patrones alternativos de empalme.
  • Se analizaron eventos de empalme alternativos regulados por MBNL, incluidos los del factor de transcripción FOXP1.

Principales resultados:

  • MBNL1 y MBNL2 son reguladores negativos directos del empalme alternativo de exones de cassette en las células ES.
  • La eliminación de MBNL en las células diferenciadas indujo un patrón de empalme similar al de las células ES; la sobreexpresión de MBNL en las células ES promovió el empalme similar al de las células diferenciadas.
  • Las proteínas MBNL regulan un interruptor de empalme alternativo específico de la célula ES en FOXP1, un factor de transcripción que controla la pluripotencia.
  • El knockdown de MBNL aumentó la expresión génica de pluripotencia e indujo la formación de células madre pluripotentes durante la reprogramación.

Conclusiones:

  • Las proteínas MBNL juegan un papel regulador central y negativo en el mantenimiento de la pluripotencia de las células ES a través del empalme alternativo.
  • El empalme alternativo, regulado por las proteínas MBNL, es un mecanismo crítico en la pluripotencia de las células ES y la reprogramación de las células somáticas.
