Protein Modifications in the RER

Modification of secretory and transmembrane proteins entering the rough ER begins in the ER lumen. These modifications aid in protein folding and stabilize the acquired tertiary structure. Protein modifications in the rough ER co-occur at different stages of protein folding.

Sus antecedentes:

  • Los enlaces disulfuro son los principales desencadenantes de respuesta a la reducción utilizados en las tecnologías actuales de administración de fármacos.
  • Los sistemas basados en disulfuro existentes tienen limitaciones en la accesibilidad sintética y la atonabilidad cinética.

Objetivo del estudio:

  • Introducir y caracterizar el tioester como una nueva química sensible al tiol para aplicaciones avanzadas de administración de fármacos.
  • Explorar la reactividad sintonizable de los tioesters mediante la modificación de su entorno estérico.
  • Desarrollar un reactivo de PEGilación de proteínas sin rastro y reversible que utilice la química del tioester.

Principales métodos:

  • Síntesis y caracterización de compuestos tioéster con diferentes entornos estéricos.
  • Estudios cinéticos para evaluar la reactividad y la sintonizabilidad de los enlaces tioesteros.
  • Desarrollo y prueba de un nuevo reactivo de PEGilación de proteínas basado en la química del tioester.

Principales resultados:

  • Los tioésteres demuestran una síntesis fácil y un orden de magnitud más amplio de afinidad cinética en comparación con los disulfuros.
  • La reactividad de los tioesters se puede modular con precisión mediante el ajuste de la masa estérica alrededor del enlace tioester.
  • Se desarrolló con éxito un nuevo reactivo de PEGilación de proteínas sin rastro y reversible utilizando la química del tioester.

Conclusiones:

  • La química de Thioester representa una plataforma versátil y altamente sintonizable para el desarrollo de la próxima generación de sistemas de administración de fármacos sensibles a la reducción.
  • La fácil síntesis y el amplio control cinético ofrecidos por los tioesters superan las limitaciones de los enfoques actuales basados en disulfuros.
  • El reactivo de PEGilación desarrollado a base de tioester es prometedor para estrategias avanzadas de bioconjugación.
