Proteins are chains of amino acids linked together by peptide bonds. Upon synthesis, a protein folds into a three-dimensional conformation, critical to its biological function. Proteins are one of the most abundant organic molecules in living systems and have the most diverse range of functions of all macromolecules. Proteins may be structural, regulatory, contractile, or protective. They may serve in transport, storage, or membranes; or they may be toxins or enzymes. Protein Organization

Proteins are polymers of amino acid residues. They are versatile and responsible for different cellular functions, including DNA replication, molecular transport, catalysis, and structural support. Proteins have a hierarchical structure comprising at least three levels of organization: primary, secondary, and tertiary structure. Some large proteins have a quaternary structure where individual protein subunits are linked together.
The primary structure of a protein is its amino acid sequence.

Objetivo del estudio:

  • Desarrollar una estrategia basada en la secuencia para crear foldameres sintéticos que imiten las estructuras terciarias de proteínas naturales.
  • Para diseñar oligómeros de columna vertebral no naturales capaces de plegarse en formas tridimensionales complejas, parecidas a las proteínas.
  • Demostrar la viabilidad de convertir proteínas naturales en análogos con columnas vertebrales parcialmente antinaturales mientras se mantiene el comportamiento plegable.

Principales métodos:

  • Utilizando un enfoque de diseño basado en secuencias para guiar el plegamiento de oligómeros espina dorsal no naturales.
  • Modificar la secuencia de una proteína natural para incorporar aproximadamente el 20% de bloques de construcción no naturales.
  • Analizando el comportamiento de plegamiento y la similitud estructural de la proteína modificada análoga a la proteína madre.

Principales resultados:

  • Diseñó con éxito un enfoque basado en secuencias para crear foldameres con estructuras terciarias similares a las proteínas.
  • Desarrolló un análogo de proteína con una columna vertebral que comprende ~ 20% de bloques de construcción antinaturales.

Conclusiones:
