Sus antecedentes:

  • Los ARN largos no codificantes (ARNlnc) son cada vez más reconocidos por su papel en los procesos biológicos.
  • Sin embargo, sus funciones específicas en la diferenciación y actividad de las células inmunes siguen siendo en gran medida inexploradas.
  • Comprender la participación del lncRNA es clave para descifrar la compleja regulación inmune.

Objetivo del estudio:

  • Identificar y caracterizar nuevos lncRNAs involucrados en la diferenciación y función de las células dendríticas convencionales (DC).
  • Para dilucidar los mecanismos moleculares por los cuales los lncRNAs identificados regulan la actividad DC.
  • Explorar el potencial de los lncRNAs como reguladores de las respuestas de las células inmunes.

Principales métodos:

  • Identificación de lnc-DC exclusivamente en DC convencionales para humanos.
  • Experimentos de knockdown in vitro en monocitos humanos y estudios in vivo utilizando células de médula ósea de ratón.
  • Análisis de ensayos de activación de células T.
  • Investigación de las vías de activación de STAT3 (transductor de señal y activador de la transcripción 3), incluyendo ensayos de unión a proteínas y fosforilación.

Principales resultados:

  • lnc-DC fue identificado como un nuevo lncRNA expresado específicamente en DC convencionales humanos.
  • La eliminación de lnc-DC deterioró significativamente la diferenciación de DC y redujo su capacidad para estimular la activación de las células T.
  • lnc-DC se une directamente a STAT3 en el citoplasma, promoviendo su fosforilación y evitando la desfosforilación por SHP1, activando así la señalización de STAT3.

Conclusiones:

  • lnc-DC es un regulador crítico de la diferenciación y función de las células dendríticas.
  • El estudio revela un nuevo mecanismo de acción del lncRNA que implica la modulación de la señalización STAT3.
  • Este hallazgo amplía las funciones conocidas de los lncRNA en la inmunidad y proporciona nuevos conocimientos sobre la regulación de las células inmunes.
