DNA replication is a well-evolved process that copies millions of base pairs with high fidelity during each cell division. Occasionally a wrong base or a long stretch of wrong bases may get added to the daughter strands. If the errors are left unchecked, cells might accumulate several mutations that might endanger their  survival. Therefore, the copying errors are checked and repaired at three levels.

Sus antecedentes:

  • Los plásmidos de estafilococos utilizan ARN antisentido para el control de la replicación.
  • Los ARN antisenso (contratranscripciones) bloquean la expresión de la proteína iniciadora.
  • pT181 es un plásmido modelo para el estudio de la regulación antisense mediada por ARN.

Objetivo del estudio:

  • Para dilucidar el mecanismo por el cual las contratranscripciones de pT181 regulan la expresión de la proteína iniciadora.
  • Para investigar el papel de las estructuras secundarias de ARNm en la regulación génica mediada por ARN antisense.
  • Para diferenciar el sistema de atenuación pT181 de los atenuadores clásicos basados en el estancamiento ribosomal.

Principales métodos:

  • Análisis de la formación de la estructura secundaria del ARNm.
  • Investigando la interacción entre las contratranscripciones y el ARNm objetivo.
  • Comparando la atenuación antisense impulsada por ARN con la atenuación inducida por tRNA.

Principales resultados:

  • Las contra-transcripciones de pT181 inducen la terminación prematura del ARNm (atenuación).
  • Esta terminación está mediada por la promoción de una estructura de horquilla cerca del codón de inicio del iniciador.
  • Una secuencia de "preemptor" aguas arriba normalmente impide la terminación al interactuar con la horquilla del terminador.

Conclusiones:

  • El plásmido pT181 emplea un nuevo mecanismo de atenuación basado en ARN antisense para el control de la replicación.
  • Este sistema difiere fundamentalmente de los atenuadores tradicionales que se basan en el retraso de los ribosomas.
  • Los ARN antisenso ofrecen una estrategia única para regular la expresión génica en los plásmidos bacterianos.
