Resumen

La adaptación del virus de la inmunodeficiencia humana tipo 1 (VIH-1) en macacos le permitió superar las restricciones del huésped y causar SIDA. La modulación inmune temprana alteró drásticamente la progresión de la enfermedad, destacando los factores en la patogénesis del SIDA.

Sus antecedentes:

  • Los lentivirus de primates tienen un estrecho tropismo de huésped, lo que limita las zoonosis pero dificulta el desarrollo de modelos animales de SIDA.
  • Comprender las barreras de transmisión entre especies del VIH-1 es crucial para desarrollar intervenciones y modelos efectivos.

Objetivo del estudio:

  • Investigar los factores que limitan la transmisión entre especies del virus de la inmunodeficiencia humana tipo 1 (VIH-1).
  • Adaptar el VIH-1 en un modelo de macaco para superar los factores de restricción del huésped y estudiar la patogénesis del SIDA.

Principales métodos:

  • La transmisión de un VIH-1 modificado en macacos de cola de cerdo con agotamiento celular transitorio de CD8 (((+) durante la infección aguda.
  • Monitoreo de la adaptación viral, los niveles de replicación y las respuestas inmunitarias del huésped en cuatro pasajes.

Principales resultados:

  • El VIH-1 se adaptó para antagonizar la teterina del macaco, un factor de restricción clave.
  • El VIH-1 adaptado se replicó en niveles más altos, causando un agotamiento significativo de las células T CD4 ((+)) y condiciones que definen el SIDA.
  • El agotamiento transitorio de las células CD8 ((+)) durante la infección aguda condujo a una rápida progresión del SIDA, a diferencia de los controles no tratados que exhiben un fenotipo de controlador de élite.

Conclusiones:

  • Una cepa adaptada del VIH-1 puede inducir el SIDA en macacos, lo que demuestra el potencial para la zoonosis.
  • Las perturbaciones inmunológicas tempranas, como el agotamiento de las células CD8 ((+), influyen profundamente en la progresión y el resultado de la enfermedad por VIH-1.
