Resumen

Las células de mamíferos utilizan la lectura translacional programada (PTR) para crear nuevas isoformas de proteína VEGF-Ax a partir del ARNm del factor de crecimiento endotelial vascular A (VEGFA). Este mecanismo recién descubierto revela un factor antiangiogénico único con implicaciones significativas en los tejidos humanos y las enfermedades.

Objetivo del estudio:

  • Para investigar la lectura translacional programada (PTR) en las células de mamíferos.
  • Para identificar nuevas isoformas de proteínas generadas por PTR y sus funciones.

Principales métodos:

  • Análisis del ARNm 3' UTR de VEGFA con elementos cis de acción.
  • Identificación de las proteínas de unión al ARN involucradas en la PTR utilizando hnRNP A2/B1.1.
  • Ensayos funcionales para la actividad antiangiogénica del VEGF-Ax.
  • Análisis de todo el genoma para objetivos adicionales de lectura.

Principales resultados:

  • Se descubrió la lectura translacional programada (PTR) en las células endoteliales de mamíferos, generando VEGF-Ax a partir del ARNm de VEGFA.
  • Se identificó un elemento de acción cis en el VEGFA 3' UTR y hnRNP A2/B1 como crucial para dirigir la decodificación serina del codón de parada UGA.
  • El VEGF-Ax exhibe propiedades antiangiogénicas, en contraste con el papel proangiogénico del VEGF-A.
  • VEGF-Ax se expresa en los tejidos humanos, pero se agota en el adenocarcinoma de colon; AGO1 y MTCH2 identificados como otros objetivos de lectura.

Conclusiones:

  • Reveló un nuevo mecanismo PTR regulado por proteínas en vertebrados.
  • VEGF-Ax representa un nuevo factor antiangiogénico con roles potenciales en la salud humana y las enfermedades.
  • Se identificaron nuevos objetivos y reguladores de la lectura translacional en los sistemas de mamíferos.
