Sus antecedentes:

  • Los productos naturales de policetida poseen diversas estructuras y bioactividades.
  • Las poliquetidas sintasas de tipo I (PKS) son grandes enzimas modulares responsables de su biosíntesis.
  • Comprender la arquitectura de PKS es clave para elucidar las vías de síntesis de policeturo.

Objetivo del estudio:

  • Para determinar la estructura tridimensional de un módulo PKS de longitud completa de *Streptomyces venezuelae*.
  • Para aclarar los mecanismos de transferencia de sustrato dentro y entre los módulos PKS.
  • Establecer un nuevo modelo para el análisis estructural de los sistemas PKS.

Principales métodos:

  • Se empleó la crio-microscopía electrónica (cryo-EM) para obtener datos estructurales de alta resolución.
  • Se generaron reconstrucciones tridimensionales de resolución subnanométrica.
  • Se realizaron comparaciones estructurales con enzimas homólogas, como la sintasa de ácidos grasos de mamíferos.

Principales resultados:

  • Se reveló una arquitectura de módulo PKS única, que difiere de las sintasas de ácidos grasos de los mamíferos.
  • Una única cámara de reacción acomoda los dominios portadores intramodules.
  • Los puntos de entrada separados facilitan la entrega del sustrato intermodular, distinto del procesamiento intramodular.

Conclusiones:

  • El estudio proporciona la primera evidencia estructural para la transferencia de sustrato intramodular e intermodular en las PKS.
  • Se propone un nuevo modelo para la función de la enzima PKS y la canalización del sustrato.
  • Este trabajo permite nuevas estrategias para la disección molecular de maquinaria PKS compleja.
