Sus antecedentes:

  • Las modificaciones de la cromatina son vitales para el desarrollo, pero sus cambios dinámicos durante la diferenciación celular siguen siendo poco conocidos.
  • La hematopoyesis (formación de células sanguíneas) es un modelo bien establecido para estudiar la dinámica del desarrollo, sin embargo, los desafíos técnicos dificultan el análisis de las poblaciones de células puras.
  • Comprender la dinámica de la cromatina es clave para descifrar los procesos de desarrollo y las decisiones del destino celular.

Objetivo del estudio:

  • Para perfilar los cambios dinámicos de cuatro modificaciones clave de la cromatina a través de 16 etapas distintas de la diferenciación hematopoyética.
  • Identificar y caracterizar las regiones potenciadoras y su comportamiento dinámico a lo largo del desarrollo de las células sanguíneas.
  • Para dilucidar la red de factores de transcripción que gobierna la dinámica de la cromatina y la especificación del linaje en la hematopoyesis.

Principales métodos:

  • Desarrollo de una técnica de inmunoprecipitación de cromatina (ChIP) de alta sensibilidad y índice primero.
  • Perfilamiento de la dinámica de modificación de la cromatina a través de 16 etapas de diferenciación hematopoyética.
  • Integración de los datos del catálogo de potenciadores con los perfiles de expresión génica.

Principales resultados:

  • Identificación y caracterización dinámica de 48.415 regiones potenciadoras.
  • El descubrimiento de 17.035 potenciadores específicos de linaje se estableció de novo durante el compromiso de linaje.
  • Demostración de que la expansión del potenciador precede y predice programas de transcripción en células diferenciadas.
  • Elucidación de la red de factores de transcripción que controla la dinámica de la cromatina y la especificación del linaje.

Conclusiones:

  • El estudio proporciona un modelo completo de la dinámica de la cromatina durante el desarrollo hematopoyético.
  • El establecimiento de potenciadores dinámicos es un mecanismo clave para la especificación del linaje y el control transcripcional.
  • Los hallazgos ofrecen nuevos conocimientos sobre la regulación epigenética de las decisiones del destino celular durante el desarrollo.
