Sus antecedentes:

  • Las proteínas intrínsecamente desordenadas (IDP) carecen de estructuras 3D estables y están implicadas en varios procesos biológicos y de enfermedades.
  • Los PDI se caracterizan típicamente por conjuntos de conformadores que se interconvierten rápidamente, lo que hace que su comportamiento dinámico sea difícil de estudiar.

Objetivo del estudio:

  • Para investigar la presencia y la naturaleza de los movimientos lentos en proteínas intrínsecamente desordenadas.
  • Explorar los mecanismos físicos que subyacen a las dinámicas correlacionadas en los PDI.
  • Para determinar si las mutaciones asociadas con la enfermedad de Parkinson afectan a estos movimientos lentos.

Principales métodos:

  • Utilizó mediciones de relajación de ciclo de campo rápido para sondear la dinámica de las proteínas en varias escalas de tiempo.
  • Aplicó cálculos hidrodinámicos basados en conjuntos para modelar movimientos correlacionados.
  • Investigó la proteína intrínsecamente desordenada alfa-sinucleína y otros PDI.

Principales resultados:

  • Se demostró que la alfa-sinucleína y otros PDI exhiben reorientaciones lentas en escalas de tiempo comparables a las proteínas plegadas.
  • Se observó que estos movimientos lentos son independientes de las propensiones estructurales secundarias / terciarias y no se ven afectados por las mutaciones relacionadas con la enfermedad de Parkinson.
  • El acoplamiento hidrodinámico entre segmentos localmente rígidos se identificó como el determinante primario de la escala de tiempo de movimiento correlacionado.

Conclusiones:

  • Las dinámicas correlacionadas de largo alcance son una característica inherente de las proteínas intrínsecamente desordenadas.
  • Propuso un mecanismo físico general para los movimientos correlacionados en sistemas biomoleculares altamente flexibles.
  • Los hallazgos proporcionan nuevos conocimientos sobre los principios físicos fundamentales que rigen el comportamiento de los desplazados internos.
