Sus antecedentes:

  • La expresión génica del virus del herpes simple (HSV) implica modificaciones post-transcripcionales, incluida la poliadenilación.
  • La selección del sitio de poliadenilación es un paso regulatorio crítico en el procesamiento del ARN.
  • Comprender los factores de procesamiento del ARN viral puede revelar los mecanismos de la regulación genética viral.

Objetivo del estudio:

  • Investigar el procesamiento del ARN en los sitios poli-A del virus del herpes simple.
  • Para identificar posibles factores celulares o virales que influyen en la eficiencia de la poliadenilación del HSV.
  • Para determinar si los sitios específicos de poli (A) viral se procesan de manera diferente en las células infectadas.

Principales métodos:

  • Ensayos de procesamiento in vitro utilizando extractos nucleares de células infectadas por HSV y células infectadas por simulacro.
  • Comparación de la eficiencia de procesamiento del sitio poli (A) utilizando ARN precursores con sitios poli (A) virales en tándem.
  • Los ensayos de labilidad térmica caracterizan la actividad de procesamiento identificada.
  • Análisis del ARN viral de virus recombinantes para confirmar los hallazgos in vivo.

Principales resultados:

  • Los extractos nucleares de las células infectadas por el HSV muestran una mayor actividad de procesamiento en un sitio específico de HSV poly (A) tardío.
  • Un segundo sitio polio (A) de HSV mostró una eficiencia de procesamiento similar tanto en extractos de células infectadas como en extractos de células infectadas simuladas.
  • Se encontró que la actividad de procesamiento específica observada era térmicamente labile.
  • Los estudios in vivo con virus recombinantes corroboraron el aumento del procesamiento en el sitio poli-A viral tardío.

Conclusiones:

  • Una actividad específica y térmica presente en las células infectadas por el HSV mejora el procesamiento de un sitio poli-viral tardío.
  • Este factor juega un papel en la regulación de la eficiencia de la poliadenilación en sitios específicos de HSV poly (A).
  • Los hallazgos sugieren un mecanismo para la regulación diferencial de la expresión génica viral durante la infección por HSV.
