Resumen

Un nuevo interruptor molecular de folato-FITC permite un control preciso sobre la terapia con células T del receptor de antígeno quimérico (CAR). Sus antecedentes:

  • Las células T diseñadas por el receptor de antígeno quimérico (CAR) (CAR-Ts) muestran una potente actividad antitumoral, pero enfrentan desafíos de seguridad debido a la activación y expansión incontroladas.
  • Las terapias actuales de células CAR-T carecen de mecanismos precisos para regular la actividad de las células T y terminar las respuestas, lo que plantea riesgos de seguridad.

Objetivo del estudio:

  • Desarrollar un nuevo interruptor molecular bifuncional para redirigir y regular la actividad de las células CAR-T.
  • Mejorar la seguridad y especificidad de la inmunoterapia con células CAR-T mediante el control de las respuestas de las células T.

Principales métodos:

  • Desarrolló un interruptor bifuncional de moléculas pequeñas: folato conjugado con isotiocianato de fluoresceína (folato-FITC).
  • Se utilizan células CAR-T específicas del FITC y células tumorales sobreexpresoras del receptor de folato (FR).
  • Se evaluó la citotoxicidad, activación y proliferación de las células CAR-T en respuesta al interruptor folato-FITC y las células diana.

Principales resultados:

  • El conmutador folato-FITC redirigió con éxito las células CAR-T específicas de FITC para apuntar a las células tumorales que sobreexpresan FR.
  • El sistema demostró una alta citotoxicidad contra las células FR-positivas sin actividad contra las células FR-negativas.
  • La activación y la proliferación de las células CAR-T dependían estrictamente de la presencia de células positivas tanto para el folato-FITC como para el FR, y eran dosis-titrables.

Conclusiones:

  • El interruptor molecular folato-FITC proporciona un método controlable y específico para involucrar a las células CAR-T con tumores objetivo.
  • Este nuevo paradigma de tratamiento ofrece una estrategia potencial para mejorar el perfil de seguridad de la inmunoterapia con células CAR-T al permitir una regulación precisa de la actividad de las células T.
