Sus antecedentes:

  • Las cataratas son la principal causa de ceguera en todo el mundo, tratadas con extracción de lentes e implantes artificiales, que conllevan riesgos.
  • No se ha demostrado previamente la regeneración de lentes en humanos.
  • La reparación de tejidos basada en células madre endógenas es un objetivo clave en la medicina regenerativa.

Objetivo del estudio:

  • Para demostrar la regeneración funcional de la lente humana.
  • Identificar los factores clave para la renovación de las células madre/progenitoras epiteliales de la lente (CEL).
  • Desarrollar un nuevo enfoque quirúrgico para el tratamiento de la catarata que preserve los LEC endógenos.

Principales métodos:

  • Células madre/progenitoras epiteliales de lentes de mamíferos aisladas (CEL).
  • Pax6 y Bmi1 identificados como cruciales para la renovación del LEC.
  • Desarrolló una técnica quirúrgica mínimamente invasiva para la extirpación de cataratas conservando LEC.

Principales resultados:

  • Regeneración de lente funcional demostrada en conejos, macacos y bebés humanos con cataratas.
  • Se confirmó el requisito de Pax6 y Bmi1 para la renovación del LEC.
  • El nuevo método quirúrgico preservó al máximo las LEC y su entorno, lo que condujo a lentes regeneradas con función visual.

Conclusiones:

  • Se ha desarrollado una nueva estrategia quirúrgica para el tratamiento de la catarata, logrando la regeneración funcional del cristalino.
  • Este enfoque preserva los LEC endógenos y su entorno natural, ofreciendo una ventaja significativa sobre los métodos actuales.
  • El estudio presenta un nuevo paradigma para la regeneración de tejidos utilizando células madre endógenas.
