Objetivo del estudio:

  • Demostrar un sistema CRISPR/Cas9 inactivo para la nucleasa para la orientación del ARN programable.
  • Para permitir el seguimiento de ARN endógeno e imágenes en células vivas sin etiquetas genéticas.
  • Para investigar la localización del ARNm y la dinámica del tráfico.

Principales métodos:

  • Utilizó Streptococcus pyogenes inactivos de la nucleasa CRISPR / Cas9 (RCas9) diseñados para la unión al ARN.
  • Se emplean ARN guía (sgRNA) para programar la orientación de RCas9 hacia ARN mensajeros específicos (mRNA).
  • Se observó la localización de RCas9 y la acumulación de ARNm en células vivas utilizando microscopía y fluorescencia in situ de hibridación.

Principales resultados:

  • El RCas9 localizado en el núcleo se exportó al citoplasma al unirse a los sgRNAs dirigidos al mRNA.
  • Se observó la acumulación de ARNm de ACTB, CCNA2 y TFRC en los gránulos de ARN.
  • Se ha demostrado el seguimiento de la resolución temporal del ARNm de ACTB a los gránulos de estrés, lo que revela la dinámica del tráfico.

Conclusiones:

  • RCas9 proporciona un método programable y libre de etiquetas para rastrear el ARN endógeno en células vivas.
  • Este sistema Cas9 dirigido al ARN facilita el estudio de la localización y la dinámica del ARNm.
  • Los hallazgos establecen una nueva herramienta para la investigación de la biología del ARN análoga a las herramientas genómicas CRISPR.
