Sus antecedentes:

  • El complejo promotor de la anafase (APC/C) es esencial para la progresión del ciclo celular en eucariotas, regulando la degradación de proteínas para la mitosis y la entrada en la fase G1.
  • La actividad de APC/C está modulada por los coactivadores Cdc20 y Cdh1, cuyas interacciones están influenciadas por eventos de fosforilación.
  • Los mecanismos precisos por los cuales la fosforilación APC/C regula la unión y la actividad del coactivador siguen siendo incompletamente entendidos.

Objetivo del estudio:

  • Para aclarar la base molecular de cómo la fosforilación controla la interacción entre el APC/C y su coactivador Cdc20.
  • Investigar el papel de los sitios específicos de fosforilación APC/C en la regulación de la progresión mitótica.
  • Comprender la regulación diferencial de APC/C por los coactivadores Cdc20 y Cdh1.

Principales métodos:

  • Para determinar la base estructural de la regulación APC/C se empleó la criomicroscopia electrónica.
  • Se realizaron análisis bioquímicos para validar las implicaciones funcionales de los cambios estructurales observados.
  • Se utilizaron ensayos in vitro para estudiar los efectos de la fosforilación y los inhibidores de moléculas pequeñas sobre la actividad de APC/ C.

Principales resultados:

  • Se identificó un interruptor molecular dependiente de la fosforilación que involucra un segmento autoinhibidor de Apc1, que regula el compromiso de Cdc20 con el APC/C.
  • El reclutamiento de complejos de Cdk-ciclina, junto con Cks, a un bucle Apc3 hiperfosforilado es crítico para aliviar esta autoinhibición.
  • El inhibidor de moléculas pequeñas tosil-l-arginina metil éster inhibe selectivamente la actividad de APC/C ((Cdc20) al interactuar con los motivos de unión clave.

Conclusiones:

  • La fosforilación de la APC/C actúa como un mecanismo regulador crítico, controlando la progresión mitótica mediante la modulación de la unión y la actividad del coactivador.
  • El estudio revela la base estructural y bioquímica para la activación de APC/C por Cdc20 durante la mitosis.
