The genome refers to all of the genetic material in an organism. It can range from a few million base pairs in microbial cells to several billion base pairs in many eukaryotic organisms. Genome assembly refers to the process of taking the DNA sequencing data and putting it all back together in a correct order to create a close representation of the original genome.   • Para reemplazar las 62.214 instancias de siete codones con alternativas sinónimas.
  • Establecer un marco para la ingeniería genética sintética a gran escala.

Principales métodos:

  • Diseño computacional del genoma y recodificación de siete codones en todos los genes codificadores de proteínas.
  • Síntesis y ensamblaje químico del genoma de 3.97 megabasas.
  • Validación de genes y segmentos del genoma recodificados mediante pruebas funcionales.
  • Identificación y corrección de las excepciones de diseño letales.

Principales resultados:

  • Recodificó con éxito 62,214 instancias de codones en todo el genoma de Escherichia coli.
  • Se validó el 63% de los genes recodificados, con el 91% de los genes esenciales conservando la funcionalidad.
  • Identificado y corregido sólo 13 excepciones de diseño letales entre 2229 genes probados.
  • Se han demostrado efectos limitados en los genes esenciales después de la recodificación.

Conclusiones:

  • La recodificación del genoma es una estrategia factible para crear organismos sintéticos con funciones mejoradas.
  • El estudio proporciona un marco sólido para el diseño, ensamblaje y análisis de genomas sintéticos a gran escala.
  • Este trabajo avanza en el campo de la biología sintética al permitir la construcción de genomas con nuevos códigos genéticos.
