  • Edición del genoma CRISPR-Cas9 para validar el papel de Gata-3.

Principales resultados:

  • Se identificó un módulo genético distinto asociado con la disfunción de las células T.
  • Este módulo se encuentra aguas abajo de las metalotioneinas intracelulares que regulan el metabolismo del zinc.
  • Gata-3, un factor de transcripción de dedo de zinc, fue identificado como un regulador clave de la disfunción de las células T.
  • La edición de CRISPR-Cas9 confirmó que Gata-3 impulsa un fenotipo disfuncional en las TIL de CD8 ((+).

Conclusiones:

  • Se ha descubierto un nuevo módulo genético que regula la disfunción de las células T, relacionado con el metabolismo del zinc y Gata-3.
  • Este hallazgo proporciona un nuevo objetivo para las terapias destinadas a restaurar la función de las células T en el cáncer y las infecciones crónicas.
