Sus antecedentes:

  • La hidratación de la superficie de las proteínas es crucial para la estructura, la flexibilidad, la dinámica y la función de las proteínas.
  • Comprender la relación entre la estructura de las proteínas y la dinámica de la hidratación es un desafío.

Objetivo del estudio:

  • Caracterizar sistemáticamente la dinámica de hidratación alrededor de una proteína beta-barril, proteína de unión de ácidos grasos del hígado de rata (rLFABP).
  • Para revelar el efecto de las diferentes estructuras secundarias de proteínas (beta-sheet vs. alfa-helical) en el comportamiento del agua de hidratación.

Principales métodos:

  • Empleamos un escáner de triptófano en la superficie de la proteína, examinando 17 sitios diferentes.
  • Se utilizó espectroscopia ultrarrápida para observar la relajación del agua de hidratación y la dinámica de la anisotropía de la cadena lateral del triptófano.

Principales resultados:

  • Se observaron tres escalas de tiempo de relajación de agua de hidratación distintas (cientos de femtosegundos a cientos de picosegundos).
  • Identificó dos escalas de tiempo de relajación de la cadena lateral de triptófano distintas (de decenas a cientos de picosegundos).
  • Las dinámicas de hidratación encontradas son heterogéneas, con el agua de la capa externa relajándose más rápido (cientos de femtosegundos) y el agua de la capa interna relajándose en escalas de tiempo de picosegundos a cien picosegundos.

Conclusiones:

  • Las dinámicas de hidratación son más rápidas que las relajaciones de proteínas y conducen a fluctuaciones de proteínas a escala de picosegundos.
  • La dinámica de hidratación es generalmente más lenta alrededor de las estructuras de láminas beta en comparación con los motivos alfa-hélicos.
  • Las proteínas de hoja beta exhiben capas de hidratación más gruesas y redes de hidratación interfaciales más rígidas que las proteínas alfa helicoidales.
