Este estudio introduce una máquina molecular artificial para sintetizar péptidos β-homo (β3) utilizando aminoácidos no proteinogénicos. La máquina funciona con éxito a través de la ligadura química nativa, creando nuevos péptidos que no se pueden producir solo por medios naturales o artificiales.

Principales métodos:

  • Síntesis de una máquina molecular artificial de tres barreras basada en el rotaxano.
  • Utilizando aminoácidos β3 no proteinogénicos para la síntesis de péptidos.
  • El uso de la ligadura química nativa (NCL) para la formación de enlaces péptidos.
  • Demostrando la eliminación de la región catalizadora por las peptidasas.

Principales resultados:

  • Síntesis y operación exitosas de la máquina molecular artificial.
  • Síntesis específica de la secuencia de los péptidos β-homo (β3).
  • Demostración de la ligadura exitosa para los estados de transición de 15 y 19 miembros.
  • Generación de un péptido único mediante la acción combinada de máquinas artificiales y biológicas.

Conclusiones:

  • Las máquinas moleculares artificiales pueden realizar la síntesis específica de la secuencia de péptidos β-homo (β3).
  • Los estados de transición de ligadura desafiantes son compatibles con la traducción de información por esta máquina artificial.
  • La integración de máquinas artificiales y biológicas permite la creación de péptidos inalcanzables por cualquiera de los sistemas de forma independiente.
