Resumen
Este estudio revela la estructura 3D de la cromatina usando la tomografía ChromEM, mostrando que es una cadena desordenada que compacta el ADN de manera diferente en la interfase y la mitosis.

Sus antecedentes:
- El modelo jerárquico de la estructura de la cromatina, basado en estudios in vitro y de microscopía electrónica (EM), propone el plegamiento jerárquico de polímeros de ADN-nucleosoma en fibras más grandes y cromosomas mitóticos.
- Visualizar la cromatina en su estado nativo (in situ) ha sido un desafío, lo que limita la comprensión de su organización tridimensional (3D).
Objetivo del estudio:
- Visualizar la ultrastructura y la organización en 3D de los polímeros de cromatina, los dominios de megabases y los cromosomas mitóticos in situ.
- Desafiar y refinar los modelos existentes de plegamiento y compactación de la cromatina.
Principales métodos:
- Desarrollo de un nuevo tinte fluorescente que tiñe el ADN y mejora el contraste para EM.
- Aplicación de la tomografía ChromEM (ChromEMT) para imágenes 3D de alta resolución de la cromatina dentro de las células.
Principales resultados:
- La cromatina existe como una cadena curvilínea desordenada con un diámetro de 5 a 24 nanómetros.
- Esta cadena está repleta de distribuciones de concentración 3D variables en interfase frente a la mitosis.
- Las cadenas de cromatina exhiben diversos arreglos de partículas y patrones de flexión para la compactación estructural.
Conclusiones:
- El modelo jerárquico establecido de plegamiento de la cromatina es probablemente una simplificación excesiva.
- La compactación de la cromatina se logra a través de un mecanismo de empaque de cadena desordenado, adaptable a diferentes estados celulares.
- ChromEMT proporciona una visión sin precedentes de la organización 3D nativa de la cromatina.
