Los puntos cuánticos (QD) con conjuntos de azúcares densos sirven como potentes sondas para las interacciones entre proteínas y glicanos multivalentes. Sus antecedentes:

  • Las interacciones multivalentes proteína-carbohidrato son cruciales para el contacto inicial virus/bacterias-célula y la infección.
  • El diseño de inhibidores multivalentes efectivos se ve obstaculizado por la falta de datos estructurales sobre proteínas de superficie celular flexibles y complejas.
  • Las lectinas como DC-SIGN y DC-SIGNR juegan un papel clave en el aumento de la entrada viral para el VIH y el virus del Ébola.

Objetivo del estudio:

  • Desarrollar puntos cuánticos (QD) que muestren matrices densas de mono- / disacáridos como sondas para las interacciones multivalentes entre proteínas y glicanos.
  • Utilizar estos glycan-QD para diseccionar los modos de unión y cuantificar las afinidades de las lectinas DC-SIGN y DC-SIGNR.
  • Evaluar el potencial de los glicanos-QD como inhibidores de la entrada viral mediada por DC-SIGN.

Principales métodos:

  • Síntesis de puntos cuánticos (QD) funcionalizados con una matriz densa de mono- / disacáridos.
  • Utilizando una combinación de transferencia de energía de resonancia de Förster (FRET), mediciones de tamaño hidrodinámico y microscopía electrónica de transmisión (TEM) para analizar las interacciones QD-lectina.
  • Desarrollo de un método QD-FRET para cuantificar las afinidades de enlace entre QD y DC-SIGN/DC-SIGNR.

Principales resultados:

  • Glycan-QDs diferenció con éxito entre los modos de unión tetra, di y monovalente de DC-SIGN y DC-SIGNR.
  • Un ensayo QD-FRET reveló que DC-SIGN se une a los QD más de 100 veces más fuerte que DC-SIGNR.
  • Glycan-QDs demostró una potente inhibición de la mejora mediada por DC-SIGN de la transducción EBOV-GP, con valores de IC50 tan bajos como 0,7 nM.

Conclusiones:

  • Los QD con visualización de glicano son herramientas versátiles para diseccionar el reconocimiento complejo de proteínas multivalentes y ligandos.
  • El método QD-FRET desarrollado cuantifica con precisión las afinidades de unión y diferencia los modos de unión de la lectina.
  • Los glicanos-QD son prometedores como inhibidores efectivos de la infección viral al dirigirse a las interacciones específicas entre proteínas y glicanos a nivel celular.
