Resumen

Sus antecedentes:

  • La segmentación del eje corporal de los vertebrados en somitas y vértebras depende del reloj de segmentación.

  • No está claro si estas oscilaciones son intrínsecas a las células individuales o a un fenómeno poblacional.

Objetivo del estudio:

  • Investigar si las oscilaciones del reloj de segmentación son una propiedad de una sola célula o a nivel de población.
  • Establecer condiciones de cultivo in vitro para la observación de oscilaciones celulares estables.
  • Para explorar el papel de la detección de quórum y señales mecánicas en el desencadenamiento de estas oscilaciones.

Principales métodos:

  • Se desarrollaron condiciones de cultivo in vitro para lograr oscilaciones celulares estables.
  • Utilizó las vías de señalización Yap y Notch para desencadenar oscilaciones.
  • Manipuló señales mecánicas dependientes de Yap para alterar estados celulares.

Principales resultados:

  • Se ha demostrado que las oscilaciones del reloj de segmentación son una propiedad colectiva de las células PSM.
  • Se ha demostrado que las oscilaciones pueden desencadenarse activamente in vitro mediante una señal dinámica de detección de quórum.
  • Se confirmó que la manipulación de señales mecánicas dependientes de Yap puede cambiar las células PSM aisladas de estados quiescentes a oscilantes.

Conclusiones:

  • El reloj de segmentación funciona como un sistema excitable.
  • Las oscilaciones son un comportamiento celular colectivo, no solo una propiedad intrínseca de una sola célula.
  • Introduce un nuevo paradigma para el estudio de la dinámica de la morfogénesis de los vertebrados.
