Objetivo del estudio:

  • Para diseñar un péptido con capacidades de unión selectiva al ADN.
  • Para investigar las interacciones de ADN dependientes de los metales y reversibles.
  • Para explorar la internalización celular inducida por el metal de los péptidos.

Principales métodos:

  • Modificación de la región básica del péptido GCN4 (br) para incluir residuos de histidina (brHis2).
  • Evaluación de la afinidad y la selectividad de la unión al ADN utilizando el sitio de ADN objetivo consensuado (5'-GTCAT-3').
  • Utilizando el complejo de paladio (Pd) para promover la interacción péptido-ADN.
  • Investigando el desmontaje de complejos reversibles con un quelador de paladio.
  • Evaluación de la translocación de la membrana celular del péptido con y sin el reactivo de paladio.

Principales resultados:

  • El péptido modificado (brHis2) por sí solo no se unió a la secuencia de ADN objetivo.
  • La adición de Pd(en) Cl2 dio lugar a una alta afinidad y unión selectiva de brHis2 al ADN.
  • La interacción péptido-ADN fue reversible y se pudo cambiar varias veces utilizando complejo de paladio y quelante.
  • El tratamiento con paladio indujo una internalización celular eficiente del péptido brHis2.

Conclusiones:

  • Un péptido diseñado puede lograr la unión selectiva de la ranura principal del ADN.
  • Las interacciones dependientes del metal ofrecen un mecanismo para el control reversible de la unión al ADN.
  • El paladio puede promover la internalización de péptidos básicos en las células.
