  • Para aclarar el mecanismo por el cual KRas y Myc cooperan para impulsar la tumorigénesis.
  • Identificar las vías de señalización clave involucradas en la reprogramación estromal durante la progresión del tumor.
  • Evaluar el potencial terapéutico de dirigirse a estas vías en un modelo de cáncer de pulmón impulsado por KRas.

Principales métodos:

  • Utilizó un modelo de cáncer de pulmón de ratón con adenomas impulsados por KRasG12D.
  • Investigó los efectos de la co-activación de Myc en el crecimiento tumoral, la proliferación y la invasión.

Principales resultados:

  • La coactivación de Myc en los adenomas impulsados por KRas promueve rápidamente el desarrollo de adenocarcinomas invasivos.
  • La progresión del tumor se caracteriza por un estroma inflamatorio, angiogénico y inmunodeprimido.
  • La CCL9 y la IL-23 derivadas del epitelio se identifican como señales clave que impulsan la reprogramación estromal.
  • CCL9 media el reclutamiento de macrófagos, la angiogénesis y la exclusión de células T y B dependientes de PD-L1.
  • La IL-23 orquesta la exclusión de las células T y B adaptativas, así como las células NK.
  • El co-bloqueo de CCL9 e IL-23 inhibe efectivamente la progresión tumoral inducida por Myc.
  • La desactivación de Myc en tumores establecidos conduce a una rápida reversión de los cambios del estroma y a la regresión tumoral.
  • La regresión tumoral tras la desactivación de Myc es independiente de las células T CD4+CD8+ pero depende de las células NK.

Conclusiones:

  • Myc reprograma extensamente el estroma del tumor para crear un entorno inmunosupresor que es esencial para la progresión del tumor.
  • Dirigirse a CCL9 e IL-23 representa una estrategia terapéutica prometedora para los cánceres de pulmón impulsados por KRas.
  • La interacción entre los oncogenes y el microambiente tumoral es crítica en el desarrollo y la progresión del cáncer.
  • Las células NK juegan un papel importante en la regresión tumoral después de la retirada de la señalización oncogénica.
