Sus antecedentes:

  • El intercambio de fluoruro de azufre (SuFEx) es una técnica química de clic emergente.
  • La química SuFEx ofrece una plataforma versátil para sintetizar nuevos compuestos.
  • Su aplicación en el descubrimiento de fármacos es un área poco explorada.

Objetivo del estudio:

  • Desarrollar y validar un método in situ para la química de SuFEx en placas de 96 pozos.
  • Para permitir la síntesis rápida y el cribado biológico de derivados del arilfluorosulfato.
  • Explorar el potencial de SuFEx en la funcionalización en etapas avanzadas de fármacos contra el cáncer.

Principales métodos:

  • Desarrolló una reacción SuFEx in situ para la conversión de fenoles en sulfatos de arilofluoro.
  • Se utilizó la síntesis y el cribado automatizados para la evaluación rápida de la actividad biológica.
  • Realizó la funcionalización en etapa tardía de fármacos anticancerosos conocidos.

Principales resultados:

  • Se identificaron tres sulfatos de arilfluoro con mejor actividad de proliferación de células cancerosas.
  • El derivado de fluorosulfato de Fulvestrant mostró una mayor actividad en las células de cáncer de mama ER+.
  • El derivado de fluorosulfato de Combretastatina A4 mostró un aumento de 70 veces en la potencia contra las células de cáncer de colon resistentes a los fármacos.

Conclusiones:

  • El método SuFEx desarrollado in situ es compatible con la síntesis y el cribado automatizados.
  • Este enfoque facilita el descubrimiento de nuevos fármacos candidatos con propiedades terapéuticas mejoradas.
  • La química de SuFEx es muy prometedora para acelerar el descubrimiento y desarrollo de fármacos.
