Resumen

Una nueva investigación muestra que las células musculares no cardíacas pueden convertirse en nuevas células musculares cardíacas en corazones embrionarios, pero no en corazones adultos. Este hallazgo desafía la idea de células madre cardíacas adultas para la regeneración del corazón.

Sus antecedentes:

  • La existencia de células madre cardíacas en corazones de mamíferos adultos para la regeneración de cardiomiocitos sigue siendo un tema de debate.
  • Estudios previos han reportado poblaciones de células madre cardíacas putativas, identificadas por varios marcadores (Sca1+, Bmi1+, Isl1+, Abc2+) o que carecen de marcadores específicos.
  • No está claro si las células madre cardíacas no identificadas pueden generar nuevos cardiomiocitos en el corazón adulto.

Objetivo del estudio:

  • Investigar el potencial de los no miocitos para generar nuevos cardiomiocitos en el corazón adulto sin depender de marcadores específicos de células madre.
  • Seguir los linajes celulares in vivo para determinar el origen de nuevos cardiomiocitos durante el desarrollo embrionario, la homeostasis adulta y después del infarto de miocardio.

Principales métodos:

  • Desarrollo de un nuevo sistema de rastreo de linaje genético sin marcadores utilizando recombinasas duales.
  • Aplicación del sistema para rastrear poblaciones de no miocitos en el corazón embrionario y adulto, así como en el músculo esquelético como control.
  • Evaluación de la generación de cardiomiocitos de novo a partir de no miocitos bajo diversas condiciones fisiológicas y patológicas.

Principales resultados:

  • Los datos de mapeo del destino revelaron que los nuevos cardiomiocitos surgen de los no miocitos en el corazón embrionario.
  • No se observó evidencia de conversión de no miocitos a miocitos en el corazón adulto durante la homeostasis o después del infarto de miocardio.
  • El sistema de rastreo de linaje detectó con éxito la generación de nuevos miocitos a partir de no miocitos en el músculo esquelético después de la lesión, sirviendo como control positivo.

Conclusiones:

  • Este estudio proporciona evidencia genética in vivo de que la conversión de no miocitos a miocitos ocurre en el corazón embrionario pero no en el corazón adulto.
  • Los hallazgos desafían el potencial miogénico de las poblaciones putativas de células madre para la regeneración cardíaca en mamíferos adultos.
  • Se presenta una nueva estrategia genética para identificar células madre endógenas en otros sistemas de órganos para la reparación de tejidos.
