Objetivo del estudio:

  • Para identificar el sustrato neuronal específico para el almacenamiento de la memoria.
  • Para investigar los cambios sinápticos en las células del engrama durante la formación de la memoria.

Principales métodos:

  • Desarrollo de la técnica dual-eGRASP (reconstitución de proteínas fluorescentes verdes a través de socios sinápticos).
  • Examen de las sinapsis entre las células del engrama CA3 y CA1.
  • Evaluación del condicionamiento contextual del miedo y la potenciación a largo plazo (LTP).

Principales resultados:

  • Se observó un aumento en el número y el tamaño de las espinas dendríticas en las células engramáticas CA1 que recibieron información de las células engramáticas CA3.
  • La conectividad mejorada entre las células de engramas se correlacionó con la fuerza de la memoria en el condicionamiento contextual del miedo.
  • Las proyecciones de engramas de CA3 a CA1 ocluyeron significativamente la potenciación a largo plazo.

Conclusiones:

  • La conectividad estructural y funcional mejorada entre las células del engrama es el correlato sináptico para la formación de la memoria.
  • Los cambios sinápticos específicos en las células de engramas CA1 que reciben la entrada de las células de engramas CA3 son críticos para la codificación de la memoria.
