Resumen

Sus antecedentes:

  • El desarrollo de sondas fluorescentes ideales para la imagen de células vivas requiere brillo, baja citotoxicidad y entrega celular eficiente.
  • El diseño de fluoróforos sintéticos enfrenta desafíos para lograr todas las propiedades deseadas simultáneamente.
  • Las sondas existentes pueden presentar citotoxicidad o fotoblanqueamiento, lo que limita su utilidad en estudios a largo plazo con células vivas.

Objetivo del estudio:

  • Diseñar y sintetizar una nueva clase de sondas fluorescentes para imágenes de células vivas con mayor brillo y menor citotoxicidad.
  • Investigar las propiedades fotofísicas y la absorción celular de un nuevo ciclófano híbrido tipo caja.
  • Demostrar la eficacia de la sonda desarrollada en aplicaciones de imágenes de células vivas utilizando microscopía confocal.

Principales métodos:

  • Síntesis de un ciclófano híbrido tipo caja, ExTzBox·4X (6·4X), mediante el enlace de unidades de viologen extendido (ExBIPY) y dipiridil tiazolothiazol (TzBIPY).
  • Caracterización fotofísica que incluye la determinación del rendimiento cuántico (ΦF = 1,00 para 6·4Cl).
  • Evaluación de la citotoxicidad en macrófagos RAW 264,7 y en células vivas mediante microscopía confocal.

Principales resultados:

  • ExTzBox·4X (6·4Cl) exhibe un alto rendimiento cuántico y no es citotóxico en altas concentraciones (100 μM) debido a su estructura rígida.
  • La microscopía confocal confirmó la absorción eficiente de 64+ por los macrófagos RAW 264.7, lo que resultó en una fluorescencia brillante.
  • La sonda no mostró foto blanqueamiento o alteración del comportamiento celular, a diferencia de un compuesto de referencia acíclico menos fluorescente.

Conclusiones:

  • Las estructuras rígidas de ciclófano en forma de caja son importantes para el desarrollo de sondas altamente fluorescentes y no citotóxicas para la obtención de imágenes de células vivas.
  • ExTzBox·4X representa un avance prometedor en la tecnología de sondas fluorescentes para la investigación biológica.
  • La sonda desarrollada permite una visualización efectiva y a largo plazo de los procesos celulares sin comprometer la viabilidad celular.
