Resumen

Las proteínas NusG y RfaH universalmente conservadas regulan la transcripción bacteriana mediante la estabilización de los complejos de alargamiento de la ARN polimerasa. Los estudios estructurales revelan cómo previenen el retroceso y cómo RfaH se dirige específicamente a los sitios supresores de polaridad operon.

Sus antecedentes:

  • NusG, RfaH y Spt5 son factores esenciales de alargamiento de la transcripción conservados en todos los dominios de la vida.
  • El NusG bacteriano mejora la elongación de la ARN polimerasa (RNAP) al prevenir el retroceso y coordinar la terminación / traducción.
  • RfaH, un parálogo de NusG, actúa específicamente en los sitios del supresor de polaridad del operón (ops) para evitar la pausa.

Objetivo del estudio:

  • Aclarar los mecanismos estructurales mediante los cuales NusG y RfaH regulan los complejos de alargamiento de RNAP (CE).
  • Para entender cómo RfaH confiere especificidad a los sitios de operaciones y suprime la pausa.
  • Proporcionar información sobre la función conservada de las proteínas de la familia NusG/RfaH/Spt5.

Principales métodos:

  • Para determinar las estructuras de alta resolución se empleó la criomicroscopia electrónica de una sola partícula (crio-EM).
  • Se resolvieron las estructuras de EC que interactúan con NusG o RfaH en los sitios de operaciones.
  • Se realizó un análisis estructural comparativo para comprender la vinculación y la función de los factores diferenciales.

Principales resultados:

  • Tanto NusG como RfaH estabilizan las EC promoviendo el emparejamiento de bases del ADN aguas arriba, suprimiendo el retroceso.
  • La estructura RfaH-opsEC revela un reconocimiento específico de las horquillas de ops y una vinculación CE más estricta en comparación con NusG.
  • La unión de RfaH impide estéricamente la conformación de RNAP requerida para la pausa inducida por horquilla.

Conclusiones:

  • NusG y RfaH comparten un mecanismo común de supresión de retroceso a través de la vigilancia del ADN.
  • RfaH posee características únicas para el reconocimiento del sitio de operaciones y la estabilización EC mejorada, excluyendo NusG.
  • Los mecanismos descubiertos destacan las funciones conservadas y especializadas de estos reguladores fundamentales de la transcripción.
