The term ribozyme is used for RNA that can act as an enzyme. Ribozymes are mainly found in selected viruses, bacteria, plant organelles, and lower eukaryotes. Ribozymes were first discovered in 1982 when Tom Cech’s laboratory observed Group I introns acting as enzymes. This was shortly followed by the discovery of another ribozyme, Ribonulcease P, by Sid Altman’s laboratory. Objetivo del estudio:

  • Determinar las estructuras por microscopía criolectrónica de la Saccharomyces cerevisiae RNase P.
  • Para aclarar la base estructural de la unión y catálisis del pre-ARN t.
  • Para proporcionar una comprensión molecular de la función de la RNasa P en los eucariotas.

Principales métodos:

  • Microscopía cryoelectrónica (cryo-EM) con una resolución de 3,5 angstroms.
  • Análisis estructural de la RNasa P sola y en complejo con el pre-tRNAPhe.
